The definition of derived functors as Kan extensions doesn't explain the story: why one should consider the Kan extensions, the universal object instead of arbitrary weak ones. In the topological world, the explanation (of homotopical limits/colimits) is more unfounded: they just say that ordinary limits/colimits are not well-defined under homotopy, but they don't explain why the one they defined is a good one.
